Studies on Clonal Propagation in the Cymbidium Floral Organ Culture in Vitro


Abstract
In order to test the using for mericlone source of floral organ in orchids, floret parts of Cymbidium ¡¿ Saxanami `Harunoumi¢¥ were cultured on Murashige and Skoog¢¥s (MS) solid or liquid medium supplemented with ¥á-naphthaleneacetic acid (NAA) +6-Benzylaminopurine (BA). Column, ovary, and flower stalk were better than sepal, petal, and lip in adventitious bud formation and chlorophyll synthesis on the MS solid medium supplemented with NAA 0. 1 ppm+BA 1.0 ppm. The activity of adventitious bud formation in ovary and flower stalk remarkably increased in 2cm inflorescence and 4cm inflorescence, respectively. In both case BA caused production of many adventitious bud, but NAA inhibited adventitious bud formation. Adventitious bud formation enhanced when the cultures were subcultured on liquid medium lacking agar.
